NADPH is a cofactor essential to the numerous enzymatic steps for fatty acids, cholesterol, DNA and amino acids biosynthesis pathways. Especially, cholesterol biosynthesis also involves numerous steps requiring NADPH and hydroxymethylglutaryl-CoA (HMG-CoA) reductase, a well-known inhibition target for famous cholesterol-lowering drugs (the "statins"), also requires NADPH as an essential cofactor. NADPH, a reducing equivalent, is prerequisite to various enzymatic steps for lipid and cholesterol biosynthesis. So, We supposed that the cytosolic NADP-dependent isocitrate dehydrogenase (IDPc) which catalyzes isocitrate dehydrogenase into -ketoglutarate producing NADPH in cytosol plays a major role in obesity.

In order to inquire these hypothesis, TG Biotech generated transgenic mouse overexpressing IDPc in specific manners. The transgenic mouse showed the functional role of IDPc on adipogenesis and biosynthesis of cholesterol. Additionally, transgenic mouse were useful of animal model which had metabolic disease.
